Abstract:

Benzo(a) pyrene is a carcinigen polycyclic aromatic hydrocarbon which diffuses into the environment from combustion of organic meterials.based on various epidemiological evidences it is related to lung, skin and liver cancer.mutagenicity, and immunosuppressivety are among important biological effects of Benzo(a) pyrene.after absorbtion and distribution in the body,it undergoes epoxidation by cytochrome P-450 system in the cells.some of the electrophilic metabolites covalently bind to cellular macromolecules and in particular to DNA. its metabolites are partially detoxified conjunction to cellular glutathione (GSH),glucoronic acid, and sulphate.biological effects of B(a) P and its metabolits depent on various factors such as species, age, sex, tissue, diet, dose, genetic system, etc.the fundemental physiological difference between newborns and adults are variations in susceptibility to chemical carcinogens observed between the two age groups.in this paper we studied the level of binding of B?(A)p to calf thymus DNA and formation of DNA adduct in the incubation systems which contained adilt and newborn rat liver and skin microsomes.results indicate that in both age groups liver microsomes in comparison to skin microsomes cause a higher increase in the level of DNA addult and the ratio of DNA aduct in the liver of adults compared to animal skin was 4.2 to 1.the quatity of adduct in the incubation systems containing liver microsomes of newborn was 1-51 higher than that of skin microsomes. Moreover phenobarbiton failed to induce cytochrome p-450 and any increase in DNA adduct level of B(a)P in the newborns rat.the level of Cytochrome P-450 in adult liver was 2.86 times higher than that in newborns, liver.

MICROSOME-MEDIATED BENZO[A]PYRENE-DNA BINDING AND INHIBITION BY CYTOSOLIC FRACTIONS FROM LIVER AND SKIN OF ADULT AND WEANLING RATS

Biotransformation of benzo[a]pyrene (BaP) in the presence of microsomal fractions derived from liver and epiderm of adult and weanling rats was examined. The aim of this study was to evaluate the effect of age on the capacity of two organs in transformation of BaP. Subcellular fractions were prepared from skin and liver by ultracentrifugation and were used as the source of BaP metabolizing enzy...

Metabolic activation of cyclopenta(cd)pyrene to 3,4-epoxycyclopenta(cd)pyrene by rat liver microsomes.

Cyclopenta(cd)pyrene (CPP), a polycyclic aromatic hydrocarbon without a bay region, is a potent bacterial mutagen. The experiments reported in this paper demonstrate that the major CPP metabolite generated by liver microsomes prepared from either phenobarbital or 3-methylcholanthrene pretreated rats is the optically active trans-3,4-dihydroxy-3,4-dihydrocyclopenta(cd)pyrene. Other experiments i...

DNA adduct formation in precision-cut rat liver and lung slices exposed to benzo[a]pyrene.

Chemical-DNA adducts provide an integrated measure of exposure, absorption, bioactivation, detoxification, and DNA repair following exposure to a genotoxic agent. Benzo[a]pyrene (BaP), a prototypical polycyclic aromatic hydrocarbon (PAH), can be bioactivated by cytochrome P-450s (CYPs) and epoxide hydrolase to genotoxic metabolites which form covalent adducts with DNA. In this study, we utilize...

simulation and experimental studies for prediction mineral scale formation in oil field during mixing of injection and formation water

abstract: mineral scaling in oil and gas production equipment is one of the most important problem that occurs while water injection and it has been recognized to be a major operational problem. the incompatibility between injected and formation waters may result in inorganic scale precipitation in the equipment and reservoir and then reduction of oil production rate and water injection rate. ...

Metabolic proficiency and benzo[a]pyrene DNA adduct formation in APCMin mouse adenomas and uninvolved mucosa.

Tumour formation may involve interactions between genetic factors and environmental carcinogens. Adenoma formation in APCMin/+ mice is associated homozygous adenomatous polyposis coli (APC) gene mutation, but the effects on carcinogen susceptibility are unknown. This study tests the hypothesis that APCMin/+ adenoma formation is accompanied by changes in metabolic proficiency and carcinogen susc...
